Close the loop on substrate

The correct feed rate depends on substrate concentration and growth rate in real time. A fixed feed profile is an open-loop guess refined by hindsight.

The measurement problem

Fed-batch control is where the cost of the sampling lag is most obvious. Feed too fast and residual substrate accumulates, the organism shifts to overflow metabolism, and inhibitory by-products build up. Feed too slowly and you starve the culture and lose productivity.

With sampling every few hours, operators run conservatively and leave yield on the table on every good batch to protect against the occasional bad one.

What a live reading changes

  • Substrate, product and by-product concentrations updated continuously.
  • Feed rate driven by the measured state, delivered to the control system over OPC UA.
  • Metabolic shifts flagged by an unexpected by-product signal hours before they show in yield.
  • Harvest called on the product curve flattening rather than on the clock.
  • A continuous record of critical process parameters instead of a handful of timestamps.

Cells and solids are handled by an inline filter of 15 µm on the recirculation loop; the model is trained on real broth rather than clean standards.
