Fraction collection and pooling decisions are made on offline assay results that arrive after the fractions have been collected.
The measurement problem
Downstream processing is a sequence of separations, and each one is a decision about what to keep. Making that decision on an assay that lags the process means either pooling conservatively — and losing yield — or pooling aggressively and risking the specification.
Continuous composition on the stream turns the decision into an observation of a curve rather than a bet on where the curve probably is.
What a live reading changes
- Composition across the separation as it elutes, rather than after.
- Pooling decisions made on a measured boundary instead of a conservative margin.
- Product and contaminant tracked together where both absorb distinctly.
- The whole run recorded, so a pooling decision can be defended later.
Aqueous streams are read at a short path — 20 µm as standard — because water absorbs strongly across much of the mid-infrared.
